@text Phosphorylation of the chloroplast light-harvesting complex induces a unique, compact, helix-like structure at its N-terminus. The N-terminal domain of the phosphorylated form of the chloroplast light-harvesting complex II, from NMR of a synthetic peptide corresponding to LHC IIb of pea [2]. The phosphate group on Thr-5 makes contacts that cause the polypeptide to fold into a compact, helix-like structure that is absent from the unphosphorylated form. Such a structure is also seen, using FTIR and CD spectroscopy, in both the N-terminal domain and the whole protein - provided the phosphate group is present [2]. The domain becomes disordered when the phosphate group is removed. The unique, compact structure of the phosphorylated N-terminus resembles that of glycogen phosphorylase, and is consistent with regulation of photosynthesis by a mechanism of guided molecular recognition [3]. Kinemage 1 presents four views of the complete structure of the phosphorylated N-terminus of LHC II based on NMR. Kinemage 2 presents the same four views of the phosphorylated LHC II N-terminus, but with the alpha-carbon backbone and the side chain of the phosphorylated amino acid, Thr-5, illustrating the key role of the phosphate group in the structure. View 1 shows the overall fold. View 2 emphasises the central position of the phosphorylated side-chain of Thr-5. View 3 illustrates that the fold of the phosphorylated LHC II N-terminus is helix-like, but obviously not an alpha-helix. In fact the turn is predominantly left-handed. View 4 illustrates that the phosphate group is a key point of interactions that determine the 3-D structure: it should be expected that the dephosphorylated form has a completely different structure. This three membrane-helix structure applies to a family of light-harvesting pigment-protein complexes that bind 50 % of the chlorophyll in nature. Coordinates are available only for alpha-carbon positions of part of the LHC II polypeptide. The polypeptide is coloured white. The green rings are those assigned to chlorophyll a; the yellow rings are those assigend to chlorophyll b; the orange lines are the carotenoid, lutein. View 1, membrane normal, looks down on to the complex from the stroma side of the thylakoid membrane. The shortest membrane helix is seen end-on. View 2, membrane parallel, shows the same features as if viewed from inside the membrane. Views 3 and 4 focus on the Glutamate-Arginine ion pairs that glue the two long helices together, and provide ligands to two chlorophyll a molecules. These charged groups must provide strong ionic interactions since they are buried in the hydrophobic lipid environment. One of our research interests in Plant Cell Biology lies in the structural effect of phosphorylation of threonine in position 5 - see Kinemages 1 and 2. Kuehlbrandt is not able to resolve the N-terminal domain in this unphosphorylated LHC II complex. One of our objectives is to provide a complete explanation of the effect of phosphorylation on the structure and function of LHC II, and so understand a fundamental regulatory event in photosynthesis - and life on earth. The protein kinase that phosphorylates LHC II is controlled by redox reactions of the photosynthetic electron transport chain. This control is an example of ÒMolecular redox signallingÓ, a process that defines many aspects of the life of cells, and that may specify the location of genes found in chloroplasts and mitochondria.
